Originari di Piqua, Ohio, i fratelli Herbert, Donald, John Jr. e Harry Mills iniziano a cantare assieme negli anni ’20 nel negozio di barbiere del padre. Durante una esibizione i ragazzi iniziano ad imitare degli strumenti musicali con la voce, dirigendo il suono con le mani davanti alla bocca, cosa che diventerà una loro caratteristica. Organizzatisi, Donald diventa il cantante leader, Harry è il cantante basso e imita la tromba, Herbert imita sax, trombone e tromba, mentre John Jr. suona l’unico strumento reale, la chitarra.
Il loro diventa un incredibile sound del tutto personale e particolare che li renderà il gruppo pop maschile più popolare d’America per la durata di oltre quattro decenni. Fattisi conoscere attraverso esibizioni radiofoniche, verso la fine del 1931 iniziano ad incidere per la Brunswick e il primo disco, Tiger Rag, a gennaio 1932 è subito n.1 nella classifica Pop e disco million seller.
Un mese dopo Dinah è anch’esso n.1 Pop e grande classico poi, fino al 1934 il gruppo ottiene un’altra quindicina di presenze nella hit parade. Nel corso di quest’anno passano alla Decca e mentre pubblicano nuovo materiale le vecchie incisioni Brunswick vengono riedite sulle etichette economiche Melotone e Perfect.
Nel 1935 sono in tournée in Inghilterra quando John Jr. si ammala e muore nel gennaio 1936. John Sr. subentra allora al figlio, dedicandosi all’imitazione della tromba basso e della tuba. Il nuovo materiale Decca ha un buon riscontro di classifica ma è solo durante l’estate del 1943 che ottengono un nuovo grande successo con Paper Doll. Il disco, pubblicato nel maggio 1942, impiega oltre un anno a salire al n.1 Pop, ma vi resta per ben 12 settimane e vende oltre 6 milioni di copie.
Nell’estate 1944 You Always Hurt The One You Love è ancora un n.1 Pop, per 5 settimane, e nell’autunno del 1952 l’ultimo n.1 Pop è The Glow-Worm.
Nei primi anni ’50 i quattro abbandonano il ‘vecchio’ sound per farsi accompagnare da veri e propri strumenti, facendo covers di titoli di artisti del momento quali Charms, Clovers e Silhouettes che, a loro volta, si erano formati musicalmente con le incisioni dei Mills Brothers. John Sr. si ritira nel 1956 e gli altri proseguono come trio, registrando per la Dot dal 1958 fino a tutti gli anni ’60, ottenendo nel 1968, in piena era soul e psichedelica, le ultime presenze in classifica con Cab Driver (n.23), My Shy Violet (n.73) e The Ol’ Race Track (n.83).
Nella prima metà dei ’70 loro incisioni appaiono su Paramount e Rainwood e all’inizio dei ’90 Donald, il più giovane dei fratelli e ultimo sopravvissuto, continua ad esibirsi in duo col figlio John III.
